Triple Effect of Nonsense-Mediated mRNA Decay Inhibition as a Therapeutic Approach for Cancer

چکیده

Cancer is a complex pathology involving different genes and cellular pathways. A combination of treatments with different targets is thus required to eliminate cancer cells. This review deals with a new potential therapeutic approach: inhibiting a single RNA degradation pathway so as to inhibit tumorigenesis by acting on different cellular processes and on the expression of various genes. Also discussed is the re-expression of various genes: tumor suppressor genes harboring a nonsense mutation, NMD-silenced oncogenes, and genes inducing a specific anticancer immune response. Lastly, putative limitations of cancer treatment by NMD inhibition are addressed.

Nonsense-mediated mRNA decay among coagulation factor genes

Objective(s): Haemostasis prevents blood loss following vascular injury. It depends on the unique concert of events involving platelets and specific blood proteins, known as coagulation factors. The clotting system requires precise regulation and coordinated reactions to maintain the integrity of the vasculature. Clotting insufficiency mostly occurs due to genetically inherited coagulation fact...

Rescue of nonsense mutations by amlexanox in human cells

BACKGROUND Nonsense mutations are at the origin of many cancers and inherited genetic diseases. The consequence of nonsense mutations is often the absence of mutant gene expression due to the activation of an mRNA surveillance mechanism called nonsense-mediated mRNA decay (NMD). Strategies to rescue the expression of nonsense-containing mRNAs have been developed such as NMD inhibition or nonsen...

NMD Microarray Analysis for Rapid Genome-Wide Screen of Mutated Genes in Cancer

Gene mutations play a critical role in cancer development and progression, and their identification offers possibilities for accurate diagnostics and therapeutic targeting. Finding genes undergoing mutations is challenging and slow, even in the post-genomic era. A new approach was recently developed by Noensie and Dietz to prioritize and focus the search, making use of nonsense-mediated mRNA de...

Nonsense Suppression as an Approach to Treat Lysosomal Storage Diseases

In-frame premature termination codons (PTCs) (also referred to as nonsense mutations) comprise ~10% of all disease-associated gene lesions. PTCs reduce gene expression in two ways. First, PTCs prematurely terminate translation of an mRNA, leading to the production of a truncated polypeptide that often lacks normal function and/or is unstable. Second, PTCs trigger degradation of an mRNA by activ...

Is There a Classical Nonsense-Mediated Decay Pathway in Trypanosomes?

In many eukaryotes, messenger RNAs with premature termination codons are destroyed by a process called "nonsense-mediated decay", which requires the RNA helicase Upf1 and also, usually, an interacting factor, Upf2. Recognition of premature termination codons may rely on their distance from either a splice site or the polyadenylation site, and long 3'-untranslated regions can trigger mRNA decay....
